Cell and Tissue Biology Research is an official journal of the Turkish Histology and Embryology Association. It is an online journal publishing research articles after full peer review. All articles are published, without barriers to access, immediately upon acceptance. One volume is published every year. Each volume consists of 4 numbers published quarterly online.
Aims and Scope
Cell and Tissue Biology Research is a peer-reviewed journal that publishes original research on all aspects of anatomy, histology, cell biology and fine structure of tissues and organs on light and electron microscopical level, neuroanatomy, and morphological techniques, as well as developmental biology, focusing on morphogenesis—the study of the emergence of form during embryogenesis, mechanisms of development, differentiation, and growth in animals and plants at the molecular, cellular, and genetic levels. Published manuscript styles include original research articles, review articles, technical notes, case reports, short communications, and letters to the editor. Novel features include: full peer review, high quality of reproduction, rapid publication, no page charges, wide readership, and fast track submission.

Cell and Tissue Biology Research aims to be indexed in all major national and international databases in the neer future.

Instructıons to Authors
Cell and Tissue Biology Research publishes original research articles, review articles, technical notes, case reports, short communications, and letters to the editor within the scope of the journal. The content of the Cell and Tissue Biology Research is determined by the Editors.
The manuscript which is submitted to the journal must not contain previously published material or material under consideration for publication elsewhere, except for a preliminary report in abstract format. Accepted manuscripts become the property of Cell and Tissue Biology and may not be republished.
The decision on acceptance of manuscripts for publication in Cell and Tissue Biology Research will be made on the basis of a peer review system.
Responsibilities of the Authors
By submitting a manuscript for publication, each author acknowledges having made a substantial contribution in the conception and design of the study, the analysis and interpretation of the results, and the writing of the paper, and has approved the final submitted version of the paper. Each author thus also acknowledges responsibility for the integrity of the manuscript, assures the originality of the manuscript, and guarantees that duplicate or redundant publications or submissions have not occurred. The Editors reserve the right to request the original data obtained in the investigation. Authors are responsible for all statements made in the text.
Manuscripts and illustrations should be submitted in English, online to the editorial office. Authors should retain their own copy, as the Editor cannot accept responsibility for damage or loss of manuscripts.
The text document must be saved as Word or RTF format. Tables must be included in the text document. Figures must be saved in the formats and at the resolution indicated below (illustrations section).
The manuscript should be typed double-spaced throughout on one side of A4 paper with at least a 2.5 cm margin on all sides. Do not divide words at the end of lines. Pages should be numbered consecutively in Arabic numerals, beginning with the title page.
Prepare a cover letter and a copyright transfer form signed by all authors.
Organize the manuscript as follows: title page, abstract (including key words at the end), introduction, material and methods, results, discussion (including the conclusions), acknowledgments, references, figure legends, and tables.
Keep acronyms and abbreviations to a minimum. When an abbreviation is used, define it at first mention and follow with the abbreviation in parentheses.
Categories of Submission Review Articles
Cell and Tissue Biology Research publishes review articles on the basis of invitation by the editor(s). However, author(s) is free to suggest topics and manuscripts for publication in the journal as a rewiev article as well. The author(s) is absolutely free to design the paper. There is no limitation in the page count in this category. References, figures, and legends follow the guidelines described below under ‘Original Articles.’ The Abstract section is needed.
Original Articles
Title Page. The following information should appear: title of article; authors’ name, and last name; affiliations with complite addresses. Identify the corresponding author and provide full mailing address, phone and fax numbers, and e-mail address. Also provide a short running title (no more than 40 spaces).
Abstract. The abstract is limited to 400 words, and should describe the essential aspects of the investigation. In the first sentence state the background; in the second sentence state your specific purpose or hypothesis; in the third, fourth and fifth sentences summarize methods, results and conclusion. No references should be cited. For indexing purposes, a small number of "key words" (no more than 5) must be supplied.
Introduction. Include brief background information on what has been done in the past in this area and the importance of your investigation. End with a statement of the purpose or hypothesis of the study.
Material and Methods. This section may be divided into subsections if it facilitates reading the paper. The research design, subjects, material used, and statistical methods should be included. Do not mix results and discussion into this section. Do not include manufacturer’s names unless the specific product is important to the procedures performed, in which case the city and state or country of the manufacturer should also be given. Indicate that informed consent has been obtained from patients who participated in clinical investigations. In animal experimentation, acknowledge that ethical guidelines were followed. When appropriate, indicate that approval was obtained from the institution’s review board.
Results. This section may be divided into subsections if it facilitates reading the paper. All results based on methods must be included. If tables and graphic material will ease the understanding of the results, include them. Cite figures to illustrate the findings of the study.
Discussion. Start with limited background information and then discuss the results of the investigation in light of what has been published in the past, the limitations of your study, and potential directions for future research. In appropriate place, cite figures and graphs. Following this information, summarize the major findings of the study and their clinical usefulness. This paragraph should address the hypothesis or purpose stated earlier in the paper.
Acknowledgments. Acknowledgments should appear on a separate page. Tis section also has to include the grant information (if any) of the investigators.
References. Section must be double spaced and begin on a separate page. References to the literature should be cited in the text by the name of the author(s) followed by the year of publication. In cases in which there are
following the year to distinguish two or more papers by the same author(s) published in the same year; example (Smith, 1981a). When two or more references are included in the same bracket, they must be quoted in the chronological order; example (Smith, 1980; Bell et al., 1984). All references must be cited in the text, and all authors should be listed in references. The reference list should be in alphabetical order.
References to articles in periodical publications must include: Names and initials of all authors, year of publication, complete title of paper, name of journal (abbreviated in accordance with Index Medicus), number of volume, and first and last page numbers. Example: Morita T., Suzuki Y. and Churg J. (1973). Structure and development of the glomerular crescent. Am. J. Pathol. 72, 349-368.
References to books must include: Name and initials of authors, year of publication, full title, edition, editor, publisher, place of publication and page numbers. Example: Powell D. and Skrabanek P. (1981). Substance P. In: Gut Hormones. 2nd ed. Bloom S.R. and Polak J.M. (eds). Churchill Livingstone. Edimburgh. pp 396-401. References to URL (Web Page) must include: Name and initials of URL owners, full title of the URL, address, and accession date in pharantheses. Example: Stern M. Radial nerve entrapment. http://www.emedicine.com/orthoped/topic549.htm (accessed Dec 2005).
Tables. Each table should be given on a separate page. Each table has a short, descriptive title. Tables are numbered in the order cited in the text. Abbreviations are defined as footnotes at the bottom of each table. Tables should not duplicate data given in the text or figures.
Figures and Legends. The complete sets of original figures must be submitted. Subjects’ names must not appear on the figures. Labels should contrast well with the background. Images should be uniform in size and magnification. Illustrations should be free of all identifying information relative to the subject and institution. Line drawings should be professional in quality. Written permission for use of all previously published illustrations must be included with submission, and the source should be referenced in the legends. Written permission from any person recognizable in a photo is required. Legends must be double spaced, and figures are numbered in the order cited in the text. Submit color prints only if color is essential in understanding the material presented. Label all pertinent findings.
Illustrations should be labelled with the figure number and author's name in soft pencil on the back identifying the top edge. Photographs should be glossy bromide prints of good contrast and well matched, preferably not mounted on card. Photographs should not exceed 17.8 x 22.2 cm. The Editor reserves the right to reduce or enlarge the illustrations. Colour photographs will be allowable only in special circumstances. Line diagrams should be drawn with black ink on tracing paper or white card or supplied as glossy prints. Illustrations should be submitted protected by resistant cardboard. Apply figure numbers to the lower left-hand corner of each photograph; dry transfer lettering (such as letraset) may be used. Digital images are welcome. They must be submitted on CDROM (CD-r or CD-RW) only. We cannot use other types of disk. Images should be TIFF file format, preferentially, although other formats could be useful. Black and white figures must be at gray scale. Color figures should be preferentially in CMYK, but RGB is also allowed. Line art files must have a 500dpi resolution, while other images must have a 300dpi resolution. Supplying digital images is not a substitute for the press set of figures.
Technical Notes
While the journal encourages the submission of full-length original articles, it will consider the publication of technical notes describing the characteristics of new instruments
of methodological improvements. In addition to a title page (formatted as described above), include a summary (250 word) describing the essence of the report, an introduction (two or three sentences of background information), a description of the technique, and a discussion highlighting the educational value of the technique. References should be limited (no more than 10 preferred) to only those that give essential background material. References, figures, and legends follow the guidelines described above in ‘Original Articles.’
Short Communications
While the journal encourages the submission of full-length original articles, it will consider the publication of short communications ensuring rapid publication of new/preliminary results of unusually educational and medically important. Whole manuscript should be maximum of 3 pages, containing maximum of 1 figure, and 1 table. In addition to a title page (formatted as described above), include a summary (250 word) describing the essence of the report, an introduction (two or three sentences of background information); materails and methods, results and a discussion highlighting the educational value of the case. References should be limited (no more than 10 preferred) to only those that give essential background material. References, figures, and legends follow the guidelines described above in ‘Original Articles.’
concise case reports. These should be unusually educational and medically important. In addition to a title page (formatted as described above), include a summary (250 word) describing the essence of the report, an introduction (two or three sentences of background information); the case report (written in the past tense), and a discussion highlighting the educational value of the case. References should be limited (no more than 10 preferred) to only those that give essential background material. References, figures, and legends follow the guidelines described above in ‘Original Articles.’
Letters to the Editor
Letters to the Editor may be used to describe in an extremely brief manner either an observation of interest to our readers, an opinion relative to the Cell and Tissue Biology Research, or constructive observations or criticisms of published material. Letters should be no more than two pages and should be submitted with a brief title. A maximum of four references may be included. Letters are published at the discretion of the journal and are subject to editing.

Değerli Meslektaşlarım,
Histoloji ve Embriyoloji alanında yapılan araştırmalar güncel yöntemlerle tedavi edilemeyen hastalıkların kesin tedavilerinin mümkün olabilmesi sürecinde önemli unsurlardan biri olarak kabul edilmekte ve pre-klinik AR-GE’nin vaz geçilmez bölümünü oluşturmaktadır.
Klasik histolojiden başlayarak gelişmiş çeşitli mikroskoplarla devam eden çalışmalarımız, moleküler ve nanobiyoteknolojilere, yardımcı üreme tekniklerine, kök hücre, rejeneratif ve rekonstrüktif tıbba, biyomedikal mühendisliğine, biyoteknoloji ve biyoinformatik gibi alanlara yayılarak daha da değer kazanmıştır.
Histoloji ve Embriyoloji’nin bilim insanları ve genç araştırmacıları şimdiye kadar yaptıkları çalışmalarla evrensel bilime katkı sağlayarak bilim disiplinimizin önemini başarılı bir şekilde ortaya koymuştur.
11. Ulusal Histoloji ve Embriyoloji Kongresi siz değerli katılımcılar tarafından hazırlanan güncel çalışmaların geniş kapsamlı paylaşım ve tartışma platformunda değerlendirilmesi amacıyla düzenlenmiştir.
16-19 Mayıs 2012 tarihinde Türk Histoloji ve Embriyoloji Derneği’nin katkılarıyla Pamukkale Üniversitesi, Tıp Fakültesi Histoloji ve Embriyoloji Anabilim Dalı tarafından düzenlenen kongremizde yerli ve yabancı konuşmacıların yanı sıra kongre bilimsel kurulu tarafından seçilmiş bildiri ve poster sunumları yer alacaktır. Ayrıca Ödül Jürisi tarafından seçilen çalışmalara Kongre Bildiri Ödülleri, Prof. Dr. Yener AYTEKİN “Türkçe Bilim Dili” Ödülleri ve Pamukkale Üniversitesi 20. Kuruluş Yıldönümü Özel Ödülü verilerek genç araştırıcıların yüreklendirilmesi hedeflenmiştir.

It is a great honor for the members of the Pamukkale University School of Medicine, Department of Histology and Embryology to organize and host the 11th National Histology and Embryology Congress (with international contribution) will take place in Pamukkale University Congress Center, Denizli, Turkey from May 16 to 19, 2012.
We would like to thank the General Assembly of the Turkish Histology and Embryology Association (THED) for giving us the oppotunity to host you in our University. It is our desire to present a scientifically satisfastory Congress with conferences, panel sessions, oral and poster presentations in a friendly atmosphere while exchanging knowledge. During the Congress, updated scientific information related to the main topics of Histology and Embryology would be discussed through 20 conferences, 35 oral and 231 poster presentations.

Lykos (Çürüksu) Vadisi’nde antik dönemde tıp Celal ġimĢek
Pamukkale Üniversitesi, Fen-Edebiyat Fakültesi, Arkeoloji Bölümü, Denizli
Denizli Lykos (Çürüksu) Ovası Batı, Güney ve Ġç Anadolu‘ya geçiĢi sağlayan kavĢak noktasında yer alır. Ova‘da hiçbir bölgede olmadığı kadar sık ve büyük antik yerleĢmeler bulunmaktadır. Bunun üç ana nedeni vardır; termal su, coğrafi konum ve iklimin uygunluğu.
Vadinin Hellenistik Dönem öncesi en önemli yerleĢimi, Herodot (VII.30) ve Xenephon‘un Anabasis‘inde (I.2.4) adı geçen Kolossai antik kentidir. Hellenistik Dönem yerleĢmelerini ise Ova ortasında yer alan Laodikeia, Attouda (Sarayköy-Hisarköy), Trapezopolis (Babadağ-Bekirler köyü), Karura (Sarayköy-Tekkeköy), Büyük Menderes Nehri‘nin batı yanında Tripolis (Yenicekent), Laodikeia‘nın kuzeyinde ise Hierapolis (Pamukkale) olarak sıralanabilir.
Çürüksu (Lykos) Vadisi Afyon‘dan baĢlayan ve Aydın‘a kadar uzanan fay hattına bağlı olan termal su kaynaklarına sahiptir. Bu nedenle Çökelez Dağı ve Honaz Dağı eteklerinden Kaklık çevresine kadar olan alanlarda çok zengin ve kaliteli traverten ocakları yer alır. Termal su vadide insanlar tarafından binlerce yıldır tedavi amaçlı kullanılmıĢ ve görkemli hamam kompleksleri yapılmıĢtır.
Laodikeia antik kentinde, Hellenistik Dönem‘de bir öğrenim merkezi olan septik (kuĢkucu) filozoflar, Antiokhos ve Theiodos yetiĢmiĢtir. Tıp öğreniminin de çok önemli olduğu Laodikeia‘da, Strabon‘un (M.S. 1. yy. baĢı) zamanında Zeuxis tarafından büyük bir grup Herophileian (antik dünyanın en ünlü hekimi) tıp okulu kurulmuĢtur (Strabon XII, 8.20). ġüphesiz bu tıp okulunda hem terapiye bağlı, hem de tıbbi yöntemlerle tedaviler yapılıyordu. Antik dönemde Denizli çevresinde yer alan; Hierapolis, Laodikeia, Attouda, Karura, Herakleia Salbace ve Eumeneia kentlerinde tıp, bir bilim dalı olarak kabul edilmiĢtir.
Antik kaynaklardan Strabon (XII/8.20) M.S. 9 yılında Lycos (Çürüksu) Vadisi‘ne gelmiĢ olup, Laodikeia, Attouda ve Karura arasında büyük saygı gören Men Karou Tapınağı ve tapınak yanında yer alan önemli bir tıp okulunun varlığından söz etmiĢtir. Strabon‘a (XII/8.20) göre, bu tıp okulu Herophileion adı altında Zeuksis tarafından kurulmuĢ ve onun zamanında bu okulun yöneticisi yani baĢhekimi Aleksandros Philalethes‘tir. Yine Strabon (XII/8.20) bu okulun tıp alanında öneminin Hikesios‘un kurmuĢ olduğu Erasistrateion (ünlü hekim ve anatomi uzmanı Teos‘lu-Sığacık‘lı Eristratos) kadar önemli olduğunu bildirmiĢtir. Bu bilgiler özellikle Lykos Vadisi‘nin antik dünyada çok önemli bir tıp okuluna sahip olduğunu göstermektedir. Bölgenin en önemli antik kenti olan Laodikeia‘da (Merkez Goncalı-Eskihisar köyleri) bir tıp fakültesi yer almaktaydı ve yukarıda sözü edilen kentlerin tıp doktorları bu fakültede eğitilmekteydi. Bununla ilgili olarak kazı çalıĢmalarında birçok kabartma ve tıbbi alet ele geçmektedir. Kutsal Kitap Ġncil‘den de anladığımıza göre Laodikeia‘da göz hekimliği çok ileri gitmiĢti ve bu kente özgü göz merhemi vardı.
Laodikeia için Polemon Hanedanlığı önemli bir yer tutar. Bunlar içinde en renkli kiĢiliğe sahip olan sofist ve hatip Marcus Antonius Polemon (M.S. 88-144), Laodikeia ve Smyrna‘daki (Ġzmir) çifte ikametiyle her iki kentin siyasi ve entellektüel yaĢamında rol oynamıĢtır. Asil bir kökten gelen Polemon‘un söylev vermedeki yeteneği, Roma Ġmparatorluk Dönemi‘nde Polemon‘un ait olduğu Ġkinci Sofistik olarak bilinen edebiyata değer veren aristokrat çevrenin övgüsünü kazanmıĢtır. Lucius Flavius Philostratus‘un Sofistler‘in YaĢamları (M.S. 180–250) adlı eserinde Polemon‘un Ġmparator Antoninus Pius‘a (M.S. 138–161) (o tarihte Roma Asya eyaleti valisi) evine davetsiz girdiği için nasıl kızdığı anlatılır. Polemon, Ġmparator Trajan‘ın (M.S. 98–117) dostluğunu kazanmıĢ ve Ġmparator, Sofiste istediği her yere serbestçe gezme ayrıcalığını tanımıĢtır (Philostratus, Sofistlerin Hayatları, 25. 532). Polemon, Ġmparator Hadrian‘la (M.S. 117–138) beraber M.S. 123‘de Trakya, Lidya, Frigya, Rodos, Atina ve Ege adalarını dolaĢmıĢtır.
Polemon fizyonomi (physiognomics) konusunda ciddi incelemeler yazmıĢtır. Buna göre bir kiĢinin fiziki görünüĢünden karakteri ve sağlığı teĢhis edilebilecektir. Polemon‘un Fizyonomi (Physiognomics) adlı eserinin elimizde yalnız Arapça çevirisi bulunur. Bergama‘lı ünlü Doktor Galen (M.S. 129 ve sonra), bu garip mantığı ciddiye almamıĢsa da Polemon‘un eserinin çok geniĢ etkisi olmuĢ ve Geç Roma, Bizans ve Arap tıp kitaplarında adı sık sık geçmiĢtir. Polemon‘un tezini daha sonraki yüzyıllarda sağlık ve hastalıklar sorularına basit cevaplar arayanlar da uygulamıĢtır.
Bölgemiz içinde yer alan ve Tavas Ġlçesi, Kızılcabölük Kasabası Vakıf Köyü sınırlarında yer alan Herakleia Salbace antik kentinin baĢ tanrısı Asklepios ve karısı Hygeia‘dır. Bu da antik kentin tıp alanında ne kadar ileriye gittiğini ve antik dönemde burada yetiĢen doktorların tüm dünyada ün saldıklarını göstermektedir. Ayrıca Herakleia Salbace antik kentinde kabartmalar ve heykeller üzerinde sağlık tanrısı Asklepios ve karısı Hygeia
sıkça betimlenmiĢtir. Bunlarla ilgili olarak antik kentte bir tıp okulu vardı ve burada antik dünyanın en meĢhur hekimleri yetiĢtiriliyordu.
Antik dönemde bölgede Ģüphesiz önemli bir tıp merkezi Eumeneia (Çivril-IĢıklı kasabası) kentinde yer almaktaydı. Kent yakınında yer alan Attanassos Hieronu etrafına kurulmuĢ önemli bir tıp okulu yer almaktaydı ve tıp bir bilim olarak kabul edilmiĢtir. Attanassos Hieronu‘nda hastalar telkin ve terapi yoluyla tedavi edilirlerken, bu tıp okulunda teĢhis-tanı ve tedavi yoluyla hizmet veriliyordu.
Hierapolis (Pamukkale) antik kenti ise termal kaynaklarıyla, sıcak hamamlarıyla antik dünyada en önemli termal tedavi merkeziydi. Mezar yazıtlarından öğrenildiği kadarıyla, birçok çevre antik kentlerden insanlar buraya gelerek tedavi görmüĢler ve vasiyetleri gereği de bu antik kentte gömülmüĢlerdir. Büyük Menderes Irmağı yanında kurulmuĢ olan Tripolis antik kentinin kuruluĢ amaçlarından biri de günümüzde Yenicekent Kaplıcaları olarak bilinen termal su kaynaklarından tedavi amaçlı yararlanılmasıdır. Yine Sarayköy-Tekke Kaplıcaları olarak bilinen bölgede antik dönemde Karura adında bir kent kurulmuĢtur. Bu kentin kaplıcalarının sağlık ve tedavi amaçlı olarak kullanıldığını antik kaynaklardan Strabon (XII. 8.17) bildirmektedir.
Binlerce yıldır bölgemizde bilime dayalı tıp ve termal suya bağlı tedavi uygulanmaktadır.

BĠLĠMSEL ARAġTIRMALARIN DOĞASI ve BĠLĠM ĠNSANLARININ GÖREV ve SORUMLULUKLARI
Bilimsel araĢtırmalar; bilim insanlarının, doğaya, insana ve topluma özgü bilgileri ortaya koyma yönündeki zihinsel çabalarını ve uygulamalarını içerir. Bilim insanları, araĢtırmalarını bağımsız olarak yürütseler bile, ortaya çıkabilecek bilginin çevre ve topluma yansımasını irdelemek ve sonuçları konusunda gerekli uyarıları yapmak sorumluluğunu da taĢırlar. Bilimsel ve teknolojik geliĢmelerin günümüzde toplum ve doğa üzerinde yoğunlaĢan etkileriyle birlikte bilim insanlarının toplumsal ve etik sorumlulukları da giderek artmaktadır. Ġnsanın yaĢamını kolaylaĢtırmak için bilim ve teknoloji üretmek. Bilim insanının sorumlulukları
Bilim insanları, bilim dünyasının kendilerine duyduğu güveni koruyacak nitelik ve nicelikte araĢtırmalar yapmak zorundadırlar. Yalnızca bilim dünyasının değil, toplumun da bilim insanlarına güven ve saygı duyması çok önemlidir. Bilim topluluğu içinde kendiliğinden gerçekleĢen bir denetleme yüzyıllar boyunca bilim etiğine dayalı güven ortamını büyük ölçüde sağlamıĢtır. Ancak, bu güven ortamı özellikle son 30 yılı aĢkın bir süredir sarsıntılara uğramıĢtır. Bunun baĢlıca nedenleri Ģunlardır:
1. Bilimsel araĢtırma destekleri ve kaynakları için gereksinim giderek artmıĢ ve bu yönde bilim insanları arasındaki yarıĢma hızlanarak büyümüĢtür.
2. Yayınlar bilimsel baĢarının ölçütü olarak daha çok önem kazanmıĢ; bu da, bilim insanları üzerinde baskılar yaratmıĢtır. Bu nedenle, en kısa yoldan yeni bilimsel veri ve sonuçlara ulaĢma çabasına girilmiĢtir.
3. Bilimsel araĢtırmaların sayısının patlama ölçüsünde arttığı günümüzde, etik sorunlar da bu patlamaya paralel olarak artmıĢtır.
Biyomedikal araĢtırmalar, etik konularda hep ön planda olmuĢtur. Bunun iki nedeni vardır: Birincisi; bu çalıĢmalarda insan ve toplum doğrudan araĢtırma konusudur. Ġkincisi; yasalar hızla geliĢen biyomedikal teknolojilerin gerisinde kalmaktadır.
Geleneksel bilim anlayıĢı içinde kendiliğinden gelen düzenlemeler; ne yazık ki, günümüzde yeterli olmamaktadır. Tüm bunlar, kimi bilim insanlarının dürüstlük anlayıĢlarında, meslek normlarında ve uygulama ilkelerinde yeni arayıĢlara yol açmıĢtır. Bu geliĢmeler ve arayıĢlar, bilim insanlarının bilimsel sorumluluklarının yeniden gözden geçirilmesini gündeme getirmiĢtir.
Bilimsel dürüstlüğün temelinde güven duygusu yatar ve tüm bilimsel iliĢkilerin ve bağlantıların özünü oluĢturur. Tüm bilim insanlarının güvene ve dürüstlüğe dayalı değerleri korumaları, öncelikle yapılması gerekendir. Bilimsel araĢtırmaya katılanlar, her zaman ve ayrıcalıksız olarak aĢağıdaki temel ilkelere bağlı olmalıdırlar:
1. AraĢtırmanın tasarımı ve yürütülmesinde en yüksek mesleki standartlara sahip olmak, 2. AraĢtırmanın yapılıĢı ve bulguların analizi sırasında özeleĢtiri, dürüstlük ve açıklığı elden bırakmamak. 3. Aynı konu üzerinde araĢtırma yapmıĢ ve yapmakta olan diğer araĢtırmacılara karĢı, onların katkılarını içtenlikle ve açıkça teslim edici bir tavır içinde olmak ve bu tavırlarını makalenin yazımında da tam olarak korumak.
4. Ġçinde bulunduğu araĢtırma grubunun tüm üyelerinin de etik dıĢı davranıĢlarda bulunmasını engellemek. Bilim kurumlarının sorumlulukları
Ġstenilen düzeyde etkin bilimsel araĢtırmalar yapabilmek için, bilim insanlarının çalıĢtığı bilim kurumlarının da bu yönde kuralları, gerekli altyapıları, yaptırımları ve saydam yönetimleri olmalıdır. Bilimde etik dıĢı davranıĢlara karĢı duyarlı ve donanımlı olunmalı ve bunlara karĢı yapısal ve düzenleyici önlemler alınmalıdır. Etik ilkeler açısından sivil toplum kuruluĢlarıyla iĢbirliği yapılmasında yarar vardır.

The control of oocyte population in mammals Joshua Johnson

The key functions of the mammalian ovary are as follows. First, the ovary produces key hormones and other factors that support health and well-being, and also cyclical changes within the female reproductive tract. Second, the ovary produces mature egg cells required for the generation of offspring. Each of these functions is dependent upon the number of oocytes present in the ovary, or, the oocyte 'population.' Oocyte population is controlled by "birth" and "death" rates: the rate of the production of new oocytes and the rate of loss during follicle atresia. It is well known that the number of oocytes is highest during fetal life, and that this number drops precipitously at or near parturition. Oocyte number then steadily declines during postnatal life until ovarian function ceases (an event referred to as menopause in humans). Thus oocyte population control was thought to have two phases, a prenatal production phase, and a postnatal loss phase. Counter to this, in the last several years, multiple laboratories have presented data suggesting that instead, oogonial stem cells persist into adulthood that produce new oocytes. This may mean that the rate of oocyte death has been underestimated. Additionally, oocyte death has been thought to occur in a passive fashion, most often as a consequence of somatic follicle cell (e.g., granulosa cell) death seen during atresia. Our group has recently shown that by manipulation of the mTOR kinase pathway, living granulosa cells can be induced to actively destroy oocytes. This means that the control of the oocyte death rate is also more complex than has been appreciated. By developing a clearer picture of the key cells and pathways that control oocyte population, we may one day be able to extend the duration of ovarian function for the purposes of health and well-being, and if desired, to augment fertility.

Morphologic, genetic and molecular based selections during preimplantation embryo development: Current applications and future directions

The development of preimplantation embryos to the blastocyst stage is, at least partially, a function of autoregulation. Two distinct features of the preimplantation embryo development are activation and cleavage of the fertilized egg and differentiation of the embryonic cells into inner cell mass (ICM) and outer cell mass (trophectoderm) at the blastocyst stage. Currently, preference of multiple embryo transfer in assisted reproductive techniques (ART) laboratories is due to inadequate knowledge to determine the health embryos. So as to identify and transfer only the best, genetically normal one healthy embryo with the highest implantation potential many non-invasive and invasive techniques are in trial to use in ART laboratories. Morphological criteria consist of evaluation of cumulus cell, oocyte maturity and morphologic properties, and continue with the evaluation of pronuclear size, number and shape. Thereafter, zygote characteristics, timely cleavage staging, cytoplasmic fragmentation, cell shape and number, blastomere size, and equality of division are then evaluated. Such evaluation has a very limited potential to select the health embryos although t is the most widely used one. A crucial restriction of the morphological criteria is the time of evaluation: day 1 assessment should be performed 16-18 h after insemination or ICSI. Days 2 and 3 should be performed 33-38 h and 68-72 h after fertilization, respectively. Top-quality embryos are the ones with 4 or 5 blastomeres on day 2 and 6-8 blastomeres on day 3 after fertilization, in absence of multinucleated blastomeres, and less than 20% fragmentation. If, on day 3, embryologists selected two embryos to transfer, the accuracy that both selections were transferred on day 5 was only 23%, while the chance that only one selection was transferred was 38%, and that neither was transferred was 39%.
Invasive embryo assessment involves blastomere biopsy and genetic analysis. These techniques are also named preimplantation genetic diagnosis (PGD) and preimplantation genetic screening (PGS). PGD can be used to detect genetic abnormalities such as aneuploidy, single gene disorders, and chromosomal translocations, thereby avoiding inherited disorders. Possible biopsy techniques for PGD or PGS include biopsy on day 1, day 3, or on day 5 at blastocyst stage biopsy (trophectoderm biopsy). The disadvantages of these include biopsying of an anucleated cell, premature cell lysis, DNA degradation, loss of cell during tube transfer,
loss of nucleus during fixation onto slide or during FISH procedure, low PCR amplification efficiency (84%), relatively high allele dropout rate (5%), errors caused by chromosomal spreading, chromosomal mosaicism, FISH signal interference, and a relatively high contamination rate (per embryo tested: 5.7%). Currently, many reproductive scientists seek to develop better techniques. We will further discuss studies on molecular and genetic biomarkers that can be used to determine healthy embryo during preimplantation periods, including analysis of genomic, proteomic, metabolomic and role of embryonic stress.

Üremeye yardımcı tedavi (ÜYTE) uygulamaları sırasında oosit ve embriyo kalitesinin giriĢimsel olmayan teknikler yardımıyla ortaya konabilmesi, in-vitro fertilizasyon (IVF) çalıĢmalarının geleceğinde çok önemli yer tutacaktır. Ġnfertilite tedavisi için ÜYTE laboratuvarlarında çalıĢan embriyologlar uzun zamandır oosit ve embriyo kalitesini ortaya koyabilmek için morfolojik değerlendirme kriterlerinden yararlanmaktadırlar. Ġmplantasyon potansiyeli en yüksek olan embriyonun seçiminde morfoloji değerlendirmesi günümüzde de çok önemlidir ve incelemeler Hoffman modulasyonuna sahip standart ıĢık mikroskoplar ile yapılmaktadır. Bu mikroskoplara optik ve dijital sistem eklentileri yapılarak polarize mikroskop özellikleri kazandırılabilmektedir (PolScope). Oositte polarize ıĢıma özelliği gösteren mayoz mekiği ve zona pellusida PolScope yardımıyla görüntülenebilmektedir. Bu yöntemle oosite ve mayoz mekiğine zarar vermeden manipülasyon, inceleme ve çeĢitli ölçümler yapmak olasıdır.
Oositin fertilizasyondan sonraki hücre bölünmeleri için mayoz mekiği son derece önemli bir rol üstlenmektedir. Oositteki mayoz mekik anomalileri fertilizasyon kusurlarına, anöploidilere ve implantasyon baĢarısızlıklarına neden olabilmektedir. Günümüzde ÜYTE laboratuvarlarında inseminasyon tekniği olarak çoğunlukla mikroenjeksiyon yönteminin kullanılması nedeniyle bu iĢlem sırasında mayoz mekiğinin gözlenebilmesi, değerlendirilmesi ve mekiğe zarar verilmemesi çok önemlidir. Bu düĢüncelerle, Gülhane Askeri Tıp Akademisi ÜYTE laboratuvarında 2007 Mart ayından beri 107S157 no‘lu Tübitak projesi kapsamında alınmıĢ olan PolScope (SpindleView; CRI, Woburn, MA, USA) cihazı 500‘den fazla hastada kullanılmıĢtır.
Özellikle kontrollü ovaryan hiperstimülasyon sonrasında az sayıda oosit elde edilen, zayıf cevaplı infertil hastalarda polarize ıĢık mikroskobu kullanım tekniklerinin doğru uygulanması ile; oosit değerlendirmesi, ICSI zamanlamasının tayini ve mayoz mekiğinin korunarak ICSI yapılması hasta yararına olacaktır. Bu analiz yönteminin kullanılması, IVF laboratuarında mikroenjeksiyon yapan embriyoloğun tıp etiğindeki ―primum nil nocere – önce zarar verme‖ temel ilkesine bağlı kalmasını ve çalıĢmalarında kendisini vicdani olarak daha rahat hissetmesini sağlayabilir.

Ġmmünohistokimya yöntemleri 1980‘lerden baĢlayarak tüm dünyada yaygın olarak kullanılagelmektedir. BaĢlangıçta önemli görülen pek çok sorun ilerleyen zaman dilimlerine çeĢitli yöntemler geliĢtirilerek aĢılmıĢ ya da aĢılmaya çalıĢılmıĢtır. Bu sunumda öncelikle alanımızda karĢılan güçlüklere ve bu güçlükleri aĢmak için kullanılabilecek yöntemlere değinilmeye çalıĢılacaktır. Kısaca değinilecek ana baĢlıklar olarak, özgüllük ve güvenilirlikle ilgili sorunlar; uygun yöntem ve materyal seçimi ile ilgili konular, arĢivleme ve görüntü analiz yöntemlerinin uygun seçimi, kantitasyonla ilgili sınırlamalar, immünoelektron mikroskopide karĢılaĢılan güçlükler olarak belirlenmiĢtir. BaĢlıklarla ilgili sorunlara çözüm oluĢturmaya yardımcı eriĢilebilecek kaynakların bir listesi de sunulacaktır.
