ENDOTHELIUM IN INFLAMMATION
• Endothelium has a heterogeneous structure that shows morphological and physiological differences according to the function properties of organs and vascular places.
Endothelium is the place where many mediators participating in
inflammation are synthesized and many mediators that are synthesized
and released from different cells during inflammation are added.
Main Functions of Endothelial Cells
1. Creating a selective permeable barrier between blood and tissues
2. Regulation of vascular tonus
3. Protection and maintenance of thromboresistance
Main Functions of Endothelial Cells
4. Metabolic function
Activation: Conversion of angiotensin I into angiotensin II
Inactivation: Bradikinin, serotonin,
prostaglandins, norepinephrine and thrombinin conversion to inactive components
Lipolysis: Breaking lipoproteins into triglycerides and cholesterol
Main Functions of Endothelial Cells
5. Synthesis of soluble factor and surface molecules involved in leukocyte activation
6. The place where regulation of inflammatory and immune reactions is added.
• Important Endothelial Synthesis Products in Systemic Inflammation
-Adhesion molecules
(E-selectin, P-selectin, ICAM-1, ICAM-2, VCAM-1, PECAM-1, fibronectin)
-Proinflammatory cytokines -Superoxide anion
-Anti-inflammatory molecules -Endothelin-1
-Procoagulant factors -Anticoagulant factors
• Endothelium Response to Cytokine Group Molecules (TNF-α, IL-1 and IFN-γ)
- Adherence (E-selectin, ICAM-1, VCAM-1 expression increases) - Prokoagulation
- Secondary cytokine induction - Vasodilatation
- Vasoconstriction
- Expression of MHC molecules - Morphological changes
