PROTEIN PURIFICATION

Centrifuge

Used in many laboratories.

Centrifugation is used to separate a complex mixture.

By separating the samples with high

centrifugal force, less dense portions can be readily separated from the mixture.

(4)

Centrifuge

At the axis of rotation, more dense particles move away from the axis,

while less dense ones get close the axis.

The pellet and supernatant separated in this manner can be used in various assays.

•The process of seperation of the

proteins according to their

physicochemical properties, is

called fractionation.

•Salt, pH, heat precipitation;

protein solubility is reduced in high salt concentration. So with the addition of the right amounts of certain salts, some proteins can be selectively precipitated.

•eg - ammonium sulfate - salt precipitation
