Diphenidol inhibited sodium currents and produced spinal anesthesia

The aim of this study was to evaluate the effect of diphenidol on blocking Na+ currents and spinal anesthesia. We used the patch-clamp method to examine if diphenidol blocked Na+ currents. Lidocaine, a common used local anesthesia, was used as control. We also evaluated the potencies and durations of diphenidol and lidocaine on spinal blockades of motor function, proprioception, and nociception in rats. Lidocaine exhibited a concn.- and

state-dependent effect on tonic blockade of voltage-gated Na+ currents in mouse

neuroblastoma N2A cells (IC50 of 8.1 and 138.9 μM at holding potentials of -70 and -100 mV, resp.). Diphenidol was more potent (IC50 of 0.77 and 62.6 μM at holding potentials of -70 and -100 mV, resp.). However, unlike lidocaine, block of Na+ currents by diphenidol lacked use-dependence. We also found that diphenidol acted like lidocaine and produced

dose-related spinal blockades of motor function, proprioception and nociception. Although diphenidol had similar potencies of spinal anesthesia compared with lidocaine it produced a much longer duration of spinal blockades than lidocaine. Our results demonstrated that intrathecal diphenidol produced a long duration and similar potency on spinal

anesthesia compared with lidocaine in rats. The anesthetic effect of diphenidol could be

in part due to its blockade of Na+ currents.

這個報告主要是以 lidocaine 為 control 組,去比較 diphenidol 阻礙鈉離子電流、 抑制行為動作的 potency 和 duration結論為 diphenidol 有較好的 duration 但 potency 與 lidocaine 差不多

我在檢索的時候打了”phone”,結果跑出了一些東西,但我其實也按看不太懂那 是什麼。

心得

Scifinder

關於 scifinder,我選擇的主題是 diphenidol,當初上課在試用的時候,很快地就 選定了這個主題,原因是因為我本身是個非常容易暈車的人,而且暈車藥對我來 說完全沒有用,暑假去蘭嶼玩需要坐 3 個半小時的船,於是就到之前打工的藥局 詢問藥師,藥師就推薦我吃了這顆藥,原本在臨床上是用來治療暈眩噁心嘔吐。 結果我使用後的效果很好,雖然海上風浪很大,船上也有很多人吐,但我完全沒 有感到不舒服,從那次之後我就對這個藥印象深刻了。

使用 scifinder 找 diphenidol 查到的資料非常多,除了關於 diphenidol 的結構、性 質、合成反應外,還有許多相關研究,包括他的衍生物特性(標題:

Diphenidol-related diamines as novel muscarinic M4 receptor

antagonists)等等,所以我個人覺得 scifinder 是個非常好用的檢索系統,尤其 對於像我們這種醫學相關科系的人,他提供我們一個實用的系統可以去搜尋專業 的資料,比起直接在搜尋引擎或維基百科等等搜尋所得的資料來的更正確,不過 他似乎只能用學校圖書館的電腦才能使用,而且同時間只能有 2 個使用者登錄, 這部分對於學生來說好像有點不太方便,希望能夠開放更多的人同時上線使用這 套檢索系統!!

Innovation

其實我還是不是很了解這個要怎麼用,對於專利這部份也還不是太了解,選主題 的時候也想了好久,隨手打了個 phone 進去,然後跑出了一堆好像跟密碼還是網 路有關的文件,但我還是都看不懂他在寫什麼。 關於 innovation 這套專利檢索系統,感覺用起來有點複雜。對我來說我是覺得比 起 scifinder,innovation 比較不實用,畢竟我大概只需要查查資料就可以,但對 於有在做研究的人,innovation 似乎也蠻有用的。不過我記得上課的時候,一直 聽到來上課的老師說,這個功能不能用、那個功能也不能用,好像是因為學校沒 有買版權的關係,所以導致我們學生有很多功能都無法使用,這樣對我們來說這 套系統好像就比較不好用了,希望以後有辦法開放多一點功能,雖然聽說越多功 能好像會很貴,但我覺得如果學校老師同學真的有需要或者真的有實用的話,那 也是學校必要花的錢。
