Effects of Chicken Essence on the Antioxidative Status in Rats

本研究共分為兩個部分:第一部分為探討攝取雞精對正常大白鼠體內抗氧化狀 態之影響。實驗動物為雄性Sprague-Dawley (SD) 大白鼠,分為四組,控制組餵 予以AIN-76 為基礎的飼料,其他組則分別添加 1.5、3.0、7.5 mL/kg body weight/day 雞精量。於 0、2、4、8 週時收集血液樣本進行相關生化值之分析,並於

8 週時將大白鼠犧牲取肝臟進行分析。第二部分是利用腹腔注射四氯化碳給予

實驗動物氧化壓力,探討雞精的投與對於在氧化壓力下之大白鼠體內抗氧化狀 態是否有影響。實驗動物、分組及飼料組成份均和實驗一相同。於第四週起每週 以腹腔注射四氯化碳 (0.2 mL/100g body weight),於 0、2、4、6、8 週時收集血液樣

本進行分析,並於第8 週時將大白鼠犧牲取肝臟進行相關生化值之分析。分析項

目為抗氧化物濃度、抗氧化酵素活性、總抗氧化力、脂質過氧化物、肝功能指數。

由結果可知,雞精對於正常大白鼠之總抗氧化狀態及抗氧化酵素活性並無顯著 影響,但血漿中脂質過氧化物則有增加的趨勢。而攝取雞精可以延緩因注射四氯 化碳所帶來肝臟受損的情況,且可增加氧化壓力下之大白鼠的抗氧化酵素活性。

雖然血漿中脂質過氧化物亦有增加之趨勢,但於總抗氧化狀態與控制組無顯著 性地影響。因此,攝取雞精可提供抵抗外來壓力的保護作用,但對於增進大白鼠 體內抗氧化力方面,仍須進一步評估。

The study included two objectives: 1. The effects of chicken essence on the antioxidative status in normal rats. 2. The effects of chicken essence on the antioxidative status in stress-induced rats. The animals in both experiments were normal male Sprague-Dawleys (SD) and randomly assigned into four groups, control (AIN-76), low dose chicken essence diet (AIN-76 plus 1.5 mL/kg/ day), medium dose chicken essence diet (AIN-76 plus 3.0 mL/kg/ day) and high dose chicken essence diet (AIN-76 plus 7.5 mL/kg/ day). Blood samples were collected in week 0, 2, 4 and 8. All animals were sacrificed in week 8 to acquire liver samples. In the second experiment, to evaluate the effects of chicken essence on the antioxidative status in rats under an oxidative stress, CCl4 was injected next day after the blood samples were collected in week 4. The concentrations of antioxidants, the activities of

antioxidative enzymes, total antioxidant status (TAS), lipid peroxidation (as TBARS), GOT, and GPT were measured. Results showed that the TAS and the activities of antioxidative enzymes did not change significantly and the plasma TBARS was increased in the chicken-essence-fed groups in the first experiment. In the second experiment, the activities of antioxidative enzymes increased, which delayed the liver

damage after the rats treated with CCl4 in the group fed with chicken essence. The TAS in the rats fed with chicken essence diet did not significantly different from the control group while plasma TBARS values increased. There was no influence of chicken essence on the antioxidative status in normal rats. Under the oxidative stress from CCl4, chicken essence could provide protections from the stress. The effect of chicken essence on improving the antioxidative status in rats still need further investigation
