Fermentation Tests - Triple Sugar Iron (TSI) Agar Test

TSI agar is especially used to identify Gram (-) bacteria such as Salmonella species .

The medium contains 0.1% glucose, 1% lactose and 1%

sucrose.

It contains phenol red as pH indicator and ferric

ammonium sulphate as H

2

S formation indicator.

Thus, in the test both fermentation of three different sugars, both hydrogen sulfide formation and gas

formation can be examined.

If the medium remains completely red, there is no reaction

Note: Occasionally black color may turn yellow. In this

case, the medium should be observed periodically.
