精胺酸添加對異種移植裸鼠大腸癌細胞生長之影響

摘要:

本研究探討精胺酸 (Arginine, Arg)添加對異種移植 裸鼠大腸癌細胞生長之影響。實驗培養人類大腸直腸 腺癌細胞株SW-480,以25號針頭注射針注入100μl細 胞液 (1*106個細胞)於裸鼠背部右側皮下後隨機分成兩 組,控制組 (Control組)給予不含Arg之粉狀飼料,實驗 組 (Arg組)給予富含Arg的粉狀飼料,兩組飼料以等氮 量、等熱量配製。經五週餵食後將裸鼠犧牲,取腫瘤 細胞及脾臟分析。結果顯示在實驗後期Arg組腫瘤體積 顯著小於Control組;在腫瘤均質液中Arg組一氧化氮 (nitric oxide, NO)的濃度顯著高於Control組,而促血管 新生因子中Arg組血管內皮生長因子受器(vascular endothelial growth factor receptor, VEGF-R)、基質金 屬蛋白酶 (matrix metalloproteinae, MMP)-2、 MMP-9皆顯著低於Control組,此外Arg組胰臟內自然 殺手細胞 (natural killer cell, NK)活性顯著高於Control 組。故推測飲食中添加Arg可增加一氧化氮在體內的表 現量及自然殺手細胞活性,並減少促血管新生因子的 生 成 , 而

材料與方法:

一.實驗設計: 將人類大腸直腸腺癌細胞株SW-480以25號針頭注 射針注入100μl細胞液 (1*106個細胞)於裸鼠背部右側 皮下後,隨機分成兩組,Control組給予不含Arg之粉狀 飼料,Arg組給予富含Arg的粉狀飼料,兩組飼料以等 二.分析項目: 定期測量體重及腫瘤體積。犧牲後取腫瘤組織分析 N O 濃 度 和 促 血 管 新 生 因 子 中 血 管 內 皮 生 長 因 子 中 VEGF及VEGF-R和MMP-2、MMP-9之表現量 。取脾 臟 測 三.統計方法:

結論:
