Gerçekleştirdiğimiz çalışmada;

1. Akut yüzme egzersizi plazma leptin düzeylerinde azalmayla sonuçlandı.

2. Egzersiz yapan ve yapmayan sıçanlarda resveratrol uygulaması plazma leptin düzeylerinde bir değişikliğe yol açmadı.

3. Akut yüzme egzersizi sıçanlarda karaciğer glikojeninde azalmayla sonuçlandı. 4. Akut yüzme egzersizi yaptırılan sıçanlarda resveratrol uygulaması karaciğer glikojeninde azalmayı önleyen bir etkiyle sonuçlandı.

5. Egzersiz yaptırılmayan sıçanlarda resveratrol uygulaması karaciğer glikojeninde artışla sonuçlandı.

6. Çalışmamızın bulguları; resveratrol uygulamasının (egzersiz yapan ve yapmayan sıçanlarda) plazma leptin düzeylerinde bir değişikliğe yol açmadığını; resveratrol uygulamasının ise aynı sıçanlarda (egzersiz yapan ve yapmayan) ise karaciğer glikojeni üzerinde koruyucu etkiye sahip olabileceğini göstermektedir.

7. Bundan sonraki benzer çalışmalarda resveratrol uygulamasının karaciğerin yanı- sıra kas glikojenini, kan glukoz seviyelerini ve plazma laktat düzeylerini nasıl etkilediğinin araştırılmasını bir öneri olarak ileri sürebiliriz.

Akut Yüzme Egzersizi Yaptırılan Sıçanlarda Resveratrol Uygulamasının Kan Leptin ve Karaciğer Glikojen Düzeylerine Etkisi

Egzersizin serbest yağ asidi ve glikoz metabolizmasını artırdığı bilinmektedir. Leptinin gıda alımını azaltmak ve enerji tüketimini artırmak gibi fizyolojik etkileri göz önüne alındığında, birçok araştırıcı leptin ve egzersiz arasındaki ilişkiyi araştırmaya yönelmiştir. Bu çalışmanın amacı yüzme egzersizi yaptırılan sıçanlarda resveratrol uygulamasının plazma leptin ile karaciğer glikojen düzeylerini nasıl etkilediğini araştırmaktır.

Bu çalışma Necmettin Erbakan Üniversitesi Deneysel Tıp Araştırma ve Uygulama Merkezinden temin edilen Enstitüsünden temin edilen Wistar-Albino cinsi erişkin erkek sıçanlar üzerinde aynı merkezde gerçekleştirildi. Çalışma protokolü aynı merkezin etik kurulutarafından onaylandı. Toplam 28 adet sıçan kullanılan araştırmada hayvanlar eşit sayıda 4 gruba ayrıldı.

Grup 1, (n:7) Kontrol grubu: Hiçbir uygulamanın yapılmadığı standart diyetle beslenen grup. Grup 2, (n:7) Yüzme Grubu: Standart diyetle beslenen ve çalışmanın bitiminde 30 dakika akut yüzme egzersizi yaptırılan grup.

Grup 3, (n:7) Resveratrol Grubu: Günlük standart diyete ek olarak dört hafta boyunca içme suyu içerisinde (10 mg/kg) resveratrol verilen grup.

Grup 4, (n:7) Resveratrol Uygulanan Yüzme Grubu: Günlük standart diyete ek olarak dört hafta boyunca içme suyu içerisinde (10 mg/kg) resveratrol verilen ve çalışmanın bitiminde 30 dakika akut yüzme egzersizi yaptırılan grup.

Dört hafta süren çalışmaların bitiminde sabah 09:00-10:00 saatleri arasında dekapite edilen

hayvanlardan kan (plazma) ve karaciğer doku örnekleri alındı. Plazma leptin düzeyleri sıçan leptin kiti kullanılarak RIA yöntemiyle (ng/ml), karaciğer glikojen düzeyleri ise immünohistokimyasal yöntemle tayin edildi.

Çalışmamızda en yüksek plazma leptin düzeyleri Grup 1 ve Grup 3‟ de elde edildi (P<0.05). Grup 2 ve Grup 4‟ün plazma leptin düzeyleri birbirinden farklı değildi. Yine çalışmamızda en yüksek karaciğer glikojen seviyeleri resveratrol uygulanan Grup 3‟de (P<0.05), en düşük karaciğer glikojen düzeyleriyse grup 2 (yüzme grubu)‟de elde edildi. Kontrol (Grup 1) ve Resveratrol Uygulanan Yüzme (Grup 4) gruplarının karaciğer glikojen düzeyleri arasında önemli bir farklılık tespit edilmedi.

Çalışmamızın bulguları bir bütün olarak incelendiğinde resveratrol uygulamasının egzersiz yapan ve yapmayan sıçanlarda plazma leptini üzerinde önemli bir etksinin olmadığı, yine resveratrol uygulamasının egzersiz yapan ve yapmayan sıçanlarda karaciğer glikojen depoları üzerinde koruyucu ve/veya düzenleyici bir etkiye sahip olduğunu göstermektedir.

The Effect of Resveratrol Supplementation on Plasma Leptin and Liver Glycogen Levels in Rats with Acute Swimming Exercise

It has been known that exercise increases free fat acide and glucose metabolism. Much more researcher considered to study relation between leptin and exercise due to effect of leptin which reduce food intake and increase energy expenditure. The aim of present study was to determine how resveratrol supplementation affect plasma leptin and liver glucogen levels.

Study was conducted on male Wistar-Albino rats that obtain Necmettin Erbakan University, Experimental Medicine Reserch and Supplementation Center, mentioned center was used for experimental procedure. Study was approved ethic committe of mentioned center. Totally 28 rats were divided to 4 groups.

Group 1, (n:7) Control group: Rats were feed standart animal food.

Group 2, (n:7) Swimming group: Rats were feeded standart rat food and exposure to 30 minutes swimming exercise the end of study.

Group 3, (n:7) Resveratrol Group: Animals were feeded normal rat food plus resveratrol by drink water (10 mg/kg/day) for 4 weeks.

Group 4, (n:7) Resveratrol Plus Swimming Group: Animals were feeded normal rat food plus resveratrol by drink water (10 mg/kg/day) for 4 weeks and exposure to 30 minutes swimming exercise the end of 4 weeks.

The end of 4 weeks supplementation animals were decapited at the 09:00-10:00 a.m. and plasma and liver samples were taken for analyses. Plasma leptin was determined by rat leptin kit by RIA (ng/ml), liver glycogen by immunohistochemical procudure.

The highest plasma leptin levels were determined in the groups 1 and 3 (P<0.05). There was no differences between group 2 and 4 for leptin. The highest liver glucogen levels were determined in the group 3 (resveratrol supplemented, (P<0.05), the lowest levels were determined in the group 2 (swimming group). Liver glucogen levels in groups 1 (control) and resveratrol swimming group (group 4).

The findings of present study show that resveratrol supplementation has no affect on plasma leptin levels, however, resveratrol supplementaion has affects of saving or regulator on liver glucogen storage in rats with exercise or non-exercise
