胰酵素顆粒之制備和腸溶膜衣包覆之研究及加速試 驗之模擬探討

在第一部份實驗中,胰酵素 (Pancreatin) 為含有澱粉酵素 (Amylase) 脂肪酵素 (Lipa- se) 及蛋白質實驗對其顆粒之備制加以研究。

結果顯示,胰酵素原料以傳統涅式粒法造粒時,不加賦型劑,直接以水為粘合劑,粘 合後,以葉片造粒機造粒時 (Ram Extruder Granulator) ,有助於活性之保留,而進 一步的將傳統涅式造粒法在制程上加以改進,則能減小造粒時之機械性的破懷,而以 此法所得的酵素顆粒,乃能維持百分之九十以上之活性,將此顆粒放入流動層造粒機 (Fluid-Bed Granulator) 內,以膠溶性聚合物 (Eudragit L30 D) 作包覆研究,發現在 固定條件及包覆材料時,包覆量愈多,乾燥時明辨是間愈長,可以使膜衣之形成更完 全,在作腸溶膜衣包覆之溶離試驗時,可防止酸液進入膜內破壤酵素之活性,最後并 以本實驗所得之產品與市售品以溶離試驗作一溶離活性之比較。

第二部分為電腦模擬加速試驗,本實驗以 PASCAL 電腦程式模擬以三種溫度 (40 , 50

60 。 C) 或五種溫度 (40 , 50 , 60 , 70 , 80 。 C) 及三個月或六個月之實驗期間所 組成的加速實驗設計,由電腦依設定的相關因子常數 ( 儲架期和活化能 ) 來模擬具有實驗之差 (2.5% , 5.0% , 7.0%) 的數據,再將模擬的實驗數據應用二種盄性關系式來評估其所 得之儲架期及活化能的準確性,由此遼解二種盄性關系式在實遼應用上的可行性而由 實驗結果顯示,實驗之差的控制對預估室溫儲架期的準確度有很大的影響性,實驗設 計中的測試溫度數目的選擇和試驗期間的長短如有足夠的涵蓋範圍使藥物臺量有明顯 的變化將使預估值更超近理論值,而線性迥歸法之傳統方法是比。
