Butylidenephthalide 藥物動力學之研究

中文摘要

川芎抗痙攣主成分 Butylidenephthalide,簡稱 Bdph,已發現此藥具有對抗心絞 痛的作用。為了更進一步瞭解其口服吸收情形及藥物藥效學與藥物動力學之關係 我們進行以下之實驗。利用高效液相層析分析技術,分析家兔靜脈注射加口服 Bdph 之濃度。以正相的矽膠充填管柱(5 μm, 4×250mm) 及正己烷與氯仿 (1:1, v/v) 所組成的混合液,分別作為固定相和移動相。此外使用波長254 nm 之紫外光與 積分儀,檢測和記錄Bdph 之波峰高度。正己烷和 95﹪的乙醇(49:1, v/v) 混合液,

則作為萃取血漿中Bdph 與其內標準品 dibutylphthalate 之溶液。以上述高效液相 層析分析方法,所測得Bdph 之血中濃度達 0.01 μg/ml 。實驗是以家兔為對象,

分別予以靜脈注射10、30 和 90 mg/kg 及口服 60 mg/kg 之 Bdph,藉此得到 Bdph 在家兔體內之藥物動力學參數,由此參數我們可得知家兔對於Bdph 之吸收,分 佈及排除情形。假設為線性藥物動力學的條件下,實驗結果顯示,Bdph 靜脈注 90 mg/kg,可得其藥物動力學參數 clearance 780.8±306.9 ml/min,V ss 118638±104153 ml,T 1/2 172.4±69.8 min ,而口服 60 mg/kg,其 Bdph 之生体 可用率約17﹪,clearance 4465.9±1548.3 ml/min,V ss 13.7×10±9.5×10ml,T 1/2 210.5±145.7 min ,觀察值之 Cmax 及 Tmax 為 0.169±0.093μg/ml 和

101.3±27.5 min。至於靜脈注射 10 及 30 mg/kg 的劑量下,因為時間濃度指數圖不 能清楚見其排除曲線,所以其動力學參數未能分析,為了改進此項缺失,則有 待將來使用氣相層析分析技術,希望進一步能夠提高其敏感度,藉此求得我們 較滿意之結果。
