This variant was previously reported to be disease causing mutation in HGMD in another case. The purpose of this study is to assess the effects of sepio-lite on the proliferation, mRNA expressions of the mineralized tissue associated genes and biomineralization of cementoblasts. Immortalized mouse cementoblasts were treated with the media containing released components of sepiolite(3 days). Proliferation of the cells was evaluated using a real-time cell analyzer. Total RNA was isolated on days 3 and 6. On day 8, von Kossa stain-ing was used to evaluate mineralization. Bone sialoprotein (BSP), osteocalcin (OCN), collagen type I(COL-I), runt-related transcrip-tion factor2 (Runx2), and alkaline phosphatase (ALP) transcripts were examined using quantitative RT-PCR. Decreased proliferation was observed in the sepiolite group on days 4 and 5 when compared to control group. Whilst, BioOss and sepiolite upregulated similarly OCN, Runx2, COL-I and ALP mRNA expressions (p < 0.01) on day 3, only BioOss increased BSP transcript on day 3 when compared to control. On day 6, only sepiolite significantly up-regulated ALP and OCN mRNA expression (p < 0.01) and there was apparently higher mRNA expression for BSP, Runx2, and COL I in sepiolite group when compared to BioOss.Both sepiolite and BioOss increased similarly mineralized nodule formation. The findings of the study revealed that sepiolite may have potential as graft material in periodontol-ogy/dentistry.

Effect of Colchicine and Atorvastatin On Cell Death Mechanisms During Inflammatory Processes In Atheroschlerosis

We evaluated the effects of different doses of colchicine, ator-vastatin, and their combinations on oxidative stress, apoptosis, and autophagy in an atherosclerotic model with lipopolysaccharide-induced inflammation in endothelial-macrophage co-culture cells. HUVEC and macrophage cells were used in this study. A HUVEC–macrophage co-culture was treated with 10 ng/ml lipopolysaccharide for 24 h to establish the inflammatory effect observed in the atherosclerotic process. LPS-treated cells were incubated with various concentrations of atorvastatin, colchicine and atorvastatin–colchicine combinations. Apoptosis, autophagy, total antioxidant status (TAS), and total oxidant status (TOS) were examined from cell lysates. When TAS and TOS values of the control, LPS, and drug groups were measured at 24 and 48 h, a significant dif-ference was observed between the control, LPS, and drug groups (P values of 0.014 and 0.013, respectively). A significant difference was found when autophagy and apoptosis levels in the control, LPS, and drug groups was measured at 24 and 48 h (P = 0.015 and P = 0.013). Apoptotic activity was found to increase in a dose-dependent man-ner with both drugs, while autophagic activity was the lowest when atorvastatin was administered alone; colchicine treatment was found to decrease autophagic activity with increase in dose. https://doi.org/10.1016/j.jbiotec.2019.05.110

In vitro organogenesis and somatic embryogenesis in Seseli lehmannii Degen inflorescence culture

Seseli lehmannii Degen (Apiaceae) is a rare endemic essential oil species of the Crimean flora included in the regional Red Data Book. First biotechnology studies of this species have been started. The inflorescences from a donor plants were collected in situ during the summer period. The inflorescence explants were cultured in vitro on RG medium, containing macro- and microsalts after Murashige and Skoog with 1.0 2.0 mg/l BAP and 0.5–2.0 mg/l IAA. Induction of etiolated haploid and diploid seedlings formation and callus development was observed after 70–90 days of culture in ther-mostat at 24± 1◦C. The explants transferring to light conditions

were changed their color to bright green. In callus some antho-cyanin color zones were identified. After 2–3 weeks culture in the light, mass adventitious buds formation, single microshoots regen-eration and somatic embryogenesis were observed.
