Özet: Sağlıklı Avrupa yayın balığı (Silurus glanis) ve karabalıklarda (Clarias ıazera) bazı hematoloJik parametrelerin normal değişim aralığını belirlemek amacıyla hematokrit, hemoglobin, kırmızı ve beyaz kan hücreleri ile trombosit sa­ yılan ve toplam plazma proteini saplanmış, Winlrobe eritrosit indisi (OAH, OAHb, OAHbY) hesapıanmıştır.

Summary: In European catfish (Silurus glanis) and Alrican catlish (Clarias ıazera), hematocrit, hemoglobin con­ centralion, red blood ceııs, white blood ceııs, trombocyle counts, lolal plasma prolein were determined and Winlrobe erytrOCyle indices (MCH, MCHC, MCV) were calculaled. A normal ranges ol some hematological parameters lor he­ allhy individuals were presenled.

Giriş

Hematologieal charaeleristies are of value in assessjng condjlions ol lish health and djsease jn Ijsh (Basjo and Arago, 1988; Kocabalmaz ve Ekin­ gen, 1982; Kori, Siakpere, 1985; Tisa al al. 1983).

The applicalion of hemaıological ıechniques including measurement ol hemoglobjn con­ centraljon, hematacrjt, red blood cells (RBC) and whjte blood cells (WBC) as well as Wjnlrobe eryt­ hrocyte jndjces have valuable in determining the health ol fjsh and monitorjng stress respanses (Ezzaı at al, 1974; sıoskopl, 1993). Total plasma proıejn js also cljnically importanı in the health sta­ tus and nutrilional condillon ol lish (Fida al al, 1988; Johnslon at al, 1987).

According lo Kocabalmaz and Ekingen (1982), hematocrit, hemoglobin, RBC, WBC, trombocyte, and tolal plasma protein values jn aneslhezjed Sj­ lurus glanis were 25.0 3.3 (%), 6.4 1.0 (gldı), 1.492 0.42 (xl06/mm3), 11.1 3 (xl04/mm3), lA (0.2 (xl04/mm3) and 2.34 0.3 (g%) respeelively. For channel catfish (Ictalurus puncıatus), lotal plasma proıein, RBC, WBC, hemaloeriı and he­ moglobin were reported as 2.2-4.5 (gldı), 1.6-2.4 (miVmm3), 28.1x103/mm3, 22.7-39.3 (%) and 4-8.4 g/dl respectively (Stoskopl, 1993). Moreover, Grjzz­ Le (1977) had been determined thaı, hemaıocrit, 34.0 1.6 (%), hemoglobin 7.0 0.34 (gldı), ery­ Irocyle 2.41 0.23 (xl06/mm3), leucocyte 173 28.0 (103/mm3) for channel catfish (Ictalurus punclalus). The dillieulty of interpreting hematologieal pro­ fjle is caused by many lactors such as genetic va­ rialjon, diet composilion and some abiotie en­ vjronmenlal eondjlions (Kocabatmaz and Ekingen, 1982; Kori-Siakpere, 1985).

This descriptive study of the basic blood eha­ racleristics is a continuation and addition lo know­ ledge ol physiology of European catfish and Alrican calljsh.

Material and Method

Source ol Fish :Live specimens of Silurus gla­ nis (1530±147 g, and 53,33±2.03 cm) and Clarias lazera (588 ±101 9 and 43.17±3.35 cm) were ob­ tained Irom Çifteler-Sakaryabaşı Fish Culture Sta­ tion. Alllish were maintained in pond prior to trans­ fer lo laboralory. The range of pond water characteristics were temperature 21.5-22 oC, pH 5.5-6.5, dissolved oxygen 5.80-6.00 mgll.

The fish were fed wilh viscera of other lish. All fish was lemale and considered heallhy on the basis ol lheir appearance and the absence of ob­ vious signs ol disease.

Blood sampling : Fish were exposed to ana­ esthesia us ing Quinaldine lo facililate of handling and blood sampling. Blood samples were drawn by direct cardiac puncture into heparinized syringes.

8100d and plasma analysis : Hematocril me­ asuremenls were made immediately by drawing well-mixed samples ol blood into heparinized ca­ pillary tubes and cenlnluging 12500 rpm for 5 mi­ nutes (Robert. 1978). T olal hemoglabin was de­ termined using cyanomelhemoglobin method using Drabkins reagent (Stoskopl, 1993). Mean cor­ puscular volume (MCV), mean corpuscular he­ moglobin (MCH) and mean corpuscular he­ moglobin concentralion (MCHC) were calculated from hemoglobin, hemataerit vatues and red blood cell counts. Total plasma protein was measured based on the biuret method (Schaperclaus, 1979

Results and Discussion

The average values for each measured blood parameters of European caıfish (Silurus glanis) and Alrican catfish (Clarias lazera) were presenled in Table 1 and Table 2 respectively.

In general hematological parameters de­ termined in our study are similar to those reported for Si1urus glanis (Kocabatmaz and Ekingen, 1982), Ictalurus punctatus (Griizle, 1977) and Cla­ rias ishenensis (Kori-Siakpere, 1985).

vertheless, reucocyte values obtained in this study were 10wer than the values reported by Ko­ cabalmaz and Ekingen (1982) and Grizzle (1977). This dillerence can be atıribuled to many factors such as, species, age, diel and water conditions. Thus, the hematological characterisıics of fish can only be meaningful in their environmenl.

The presenI study was undertaken to establish a normal hematologica1 profiles of Silurus glanis and Cıadas lazera to provide a basis for com­ paratiye surveys. it is hoped that this preliminary study could conlnbule lo knowledge of physiology of these species.
