飲水中添加砷化合物對大白鼠體內氧化狀態及血壓之影響 周欣如1、楊惠婷2、黃士懿1 1台北醫學大學保健營養學研究所 2台北醫學大學藥學研究所 本研究旨在探討砷化合物甲基化路徑是否為引起周邊組織氧化壓力升高的 起始者。以wistar大白鼠分別於飲水中長期添加濃度 50 ppm 之三價砷(iAsⅢ )或 五價砷(iAsⅤ ),測試砷化合物對大白鼠體內氧化狀態及血壓影響。36 隻雌雄各半 wistar大白鼠,隨機分為 50 ppm 無機砷組((含iAsⅢ、iAsⅤ

)、正常飲水組三組, 在測得尾靜脈血壓及採尾靜脈血後進入實驗期。第一部份為氧化壓力變化,以每 隔四週採尾靜脈血進行超氧化歧化酶(SOD)、麩胱甘月太過氧化酶(GPX)與過氧化 氫酶及脂質過氧化物(MDA)含量分析。第二部分為血壓測量,以每兩週測量收縮 壓和平均血壓。結果發現,在飲水中含無機砷之組別中,SOD、GPX之酵素活 性顯著較高(p<0.05),脂質過氧化產物於四週後也有明顯增加的情形,而其增加 的程度以三價砷組較高;在飲用含無機砷水八週後,雄鼠與正常飲水組比較,其 收縮壓則明顯較高(砷水組 130 mmHg、正常組 110 mmHg,p<0.05),雌鼠仍無 顯著差異。由此發現,在砷化合物暴露下,可能藉由氧化壓力增加,使得周邊血 管造成損傷,進而使大鼠血壓上升。 關鍵詞:無機砷、氧化壓力、血壓
