PRELIMlNARY DETERMINATION OF
TRANSFER-RIN TYPES IN KONYA MERINO SHEEP

Surnrnary Blood sera of two hundred Konya merino sheep were analyzed for trans-ferrin types with horizonta\ starch g-d electrophoresis in a discontinuous buffer system. Six transfcrrin aneles, TfA, TfB, TfM, TfD, TfE and TfS, confi! IT.cd on the basis of inter-national nomenclatures, ",ere obsen'cd in the sera. Sixtecn transferrin phenotypes TfAA, TfAB, TfAM, TfAD, Tf AE, TfBB, TfBM, TfBD, TfRE, TfMM, TfMD, TfME, TfMS, TfDD, TfDE and TfEE, have been detccted in Konya merino sheep. A new trans-ferrin allele, TfS, was found in this in\"estigation.

Özet 200 adet Konya "ferinos Koyunundan alınan kan serumunda, aralıklı tampon sistem i~,inde, yatay nişasta peltesi elektroforez yiintemiyle betaglobulin (transferrin) tipleri tayini yapıldı. Muayene edilen serumlarda Uluslararası nomcnklatür asıllarına uygunlu-ğu doğrtılanan altı transferl'in allel (TfA, TfB, TfM, Tl'D, TfE ve TfS) ~aptandı. Konya Merinos Koyuıılarında TfAA, Tl'AB, TtAM, TfAD, TfAE, TfBB, TfBM, TmD, TfBE, TfMH, Tl' MD, TfME, TfMS, TfD]), TfDE, TfEE olmak üzere on altı transferrin fen 0-tip tesbit edildi. Bu araşrırmada TfS adı verilen yenibir transferrin allel bulundu.
Introduction
Sİnce the İntroductİon of starch gel electrophoresİs by Smithies in 1955 (8), the intraspecİfic polymorphisms of transferrİn (~- gl 0-bulin) İn the sera of dİfferent specİes of farm anİmals have been exa-mİned by a number of workers. of animal populations. In sheep each aııele causes the production of two zones, one densely stained and one faintly stained on starch gels. Balbiertz et al . 1968 (3); Ashton and ferguson 1962 (2) and Osman 1967 (5).
Until recently, however, sixteen sheep transferrin aııeles, TfI, Tff, TfA, TfG, TfB, TfB, TfC (TfC Hungary), TCM, TfD, TfU, TfN (TfN Hungary), TfQ, TfE, TfR, TfV and TfP have been iden-tified by a number ofworkers; Ashton 1958 (I), Fesüs 1968 (4).
The purpose of the present investigation was to present the trans-ferrin types in Konya merino sheep and to determine the distribution of individual types in a selected flock.
Materials And Methods
The sera used for the clectrophoretic separation were obtained from two hundred Konya merino shecp bred at the Konya Zooteeh. nic Research Institu te, Turkey.
Horizontal starch gel electrophoresis technique described by Smithies 1955 (8) and Poulik 1957 (7) was employed with slight modifications under the foııowing conditions:
(a) starch extracted from potatoes and hydrolyzed in the labo-ratory: 120 mL. acetone and 8 mL. hydrochlorie acid

i00 gm. of starch.(b) Electro-vessel buffer; 18.54 gm. boric acid and 2.1 gm. sodium hydroxide

i litre of distilled water.(c) Gel bufrer: 9.2 gm. Tris (hydroximethyl) - aminomethane and 1.5 gm. citric acid

from the hydrolyzed lot and the sera were inserted into the gel s on pieces of Whatman filter paper no. IIi. Ten to eleven samplcs were applied at a time and the elcctrophoresis was continued for two and a half hours at an applied voltage of 350. The gel wascut horizontaııy into slices aboLlt 2 mm. thiek and stained with Amido Black iO B solution. The bands were developedover night in the development solution containing methyl alcohol, glacial acetic acid and distiııed water.A number of transferrin types were observed on the gcls. The aııdes wc re first arbitrarily named and then were canfirmed on the basis of international nomenclatures by running them with the

rcnce sera obtained from WAGENINGEN under the similar elect-rophoretic conditions.
Results And Discussion
Six transferrin zone pairs were found to occur in the blood sera of Konya merino sheep. Starting from the fastest zone the following nomenclatures, TfA, TfB, TfM, TfD, TfE and TfS, were assigned to the alleles. Sixteen transferrin phenotypes of which five homozy-gous, TfAA, TfBB, TfMM, TfDD and TfEE and eleven heterozygous, TfAB, TfAM, TfAD, TfAE, TfBM, TfBD, TfBE, TfMD, TfME, TfMS and TfDE were identified in the investigation. The quantita-tive distribution of individual transferrin types in the fIock of sheep examined was given in the Tablc i.

Total - 200TfB, TfD and TfE alleles were not identified in Australian merino sheep whereas the presence of these alleles were detected in Polish as well as in Konya merino sheep . On the other hand Tfe allele was found in Australian, Polish and Karoo merino sheep but was absent in Konya merino sheep; Ashton and Ferguson 1962 (2), Balbiertz et aL. 1968 (3) and Osterhoff et aL. 1970 (6).
A new transferrin allele, not mentioned in the recent litera tures reviewed, was identified in Konya merino sheep. The migration rate of this allele was slower than TfE which was the slowest type of the WAGENI~GEN. Due to the decreased migratinn rate this newallele was named as TfS.
