Os cDNAs PKCI e SHAGGY estão conservados em vários grupos de plantas e parecem exercer função crucial no desenvolvimento destas;
A superexpressão do cDNA PKCI de cana-de-açúcar e a redução na expressão do gene endógeno com a construção em antissenso altera o desenvolvimento normal de plantas de tabaco transgênico;
A superexpressão do cDNA SHAGGY de cana-de-açúcar em plantas de N. tabacum parece alterar a morfologia do gineceu das flores de tabaco transgênico;
O cDNA PKCI parece estar distribuído dentro do genoma de cana-de- açúcar nas mesmas posições em ambas variedades precoce quanto das variedades tardia.
7. CONSIDERAÇÕES FINAIS
Os resultados deste trabalho consistem nos primeiros resultados da superexpressão de genes relacionados com floração em cana-de-açúcar. Até então, não existem trabalhos publicados sobre a genética molecular da floração utilizando cana-de-açúcar como alvo. Nossos resultados e os dados existentes na literatura com outras plantas modelo sugerem uma possível interação de scPKCI e scSHAGGY com outras proteínas. Essa hipótese será testada com os ensaios de duplo-híbrido em levedura. Além disso, as linhagens de plantas transgênicas para todas as construções estão sendo obtidas na progênie T3. Com esta descendência, pretende-se confirmar os fenótipos observados, assim como analisar o padrão de expressão de alguns dos genes chaves associados ao processo de florescimento nas plantas transgênicas. Tendo sido obtidas as linhagens T3, alguns ensaios fisiológicos poderão ser realizados de modo a averiguar o comportamento das plantas transgênicas em respostas a fatores ambientais e hormonais, tais como variação de fotoperíodo, aplicação de brassinosteróides, os quais influenciam no tempo de floração. Além disso, estão sendo obtidas plantas transgênicas 35S::SHAGGY/AS para analisar o efeito fenotípico da diminuição dos níveis de expressão de scSHAGGY.
As análises dos níveis de expressão dos genes scPKCI e scSHAGGY e dos genes chaves da floração nas plantas transgênicas por pela técnica de PCR em tempo real já estão sendo realizadas. Além disso, os ensaios in vivo utilizando a metodologia de duplo-híbridos estão em andamento para analisar a interação proteína-proteína para esses genes. Esses experimentos permitirão o fechamento dos resultados para a caracterização funcional destes dois cDNAs.
