Esta pesquisa demonstrou que, nas condições estudadas, o parecoxibe determinou maior quantidade de necrose, menor quantidade de apoptose e maior quantidade de células viáveis nos rins dos ratos submetidos ou não à hemorragia aguda.

Esses resultados, que podem contribuir para geração de conhecimentos relacionados a esta classe de medicamentos, prudente notar-se, também podem ter trazido novas questões.

A literatura mostra resultados conflitantes relacionados com o uso de COXIBEs e proteção renal, talvez pela diversidade metodológica. Este estudo, dentro das condições estabelecidas, sugere tendência ao benefício do uso do fármaco em relação à proteção renal.

Entretanto, esta pesquisa focou os critérios morfológicos da morte celular, e este tipo de classificação tem sido muito estudado recentemente por pesquisadores, inclusive, novas classificações sendo continuamente propostas. Desse modo, em futuro próximo, pode ser que os critérios morfológicos aqui estudados não sejam os mais acurados para entender a dinâmica da proteção relacionada com esta classe de medicamentos.

Além desta questão taxonômica, este estudo apresentou limitação quanto à determinação das vias de apoptose relacionadas com a proteção evidenciada. Isso pode ser algo sensato a ser considerado, pois, na identificação da via (e suas cascatas) podem- se criar modos de focalizar a terapia ou a proteção, sem demais prejuízos.

No sentido da anatomia, conhecem-se sítios renais específicos para as afecções – túbulos, medula, vasos e células corticais intersticiais. Neste estudo, como o rim foi visto como um todo, o método não contemplou os sítios anatômicos relacionados com os padrões morfológicos de morte celular, e esta questão também permanece aberta.

Um ponto intrigante pode ser a relação temporal entre inflamação, uso do medicamento e análise da citometria de fluxo. Será que a prevenção da apoptose significou verdadeiro salvamento da morte celular, ou será que simplesmente ocorreu um atraso da sinalização apoptótica? Esta questão prudentemente deve ser levantada.

Além de tudo, o entendimento da biologia molecular está cada vez mais presente na ciência moderna. Assim, considerar o estudo das proteínas relacionadas com dano ou proteção também pode ser algo a se pesquisar. Métodos de avaliação de proteínas, como Western Blot, ou análises de imagens de microscopia eletrônica no intuito de observar os fosfolipídeos sinalizadores de apoptose podem ser caminhos científicos futuros.

De qualquer modo, com a criação de pequenas evidências, são possíveis novas questões no sentido de trazer maiores benefícios para esta e próximas gerações, e assim a dinâmica da ciência estabelece-se e perpetua-se.

7 CONCLUSÃO

Em rins de ratos analisados por citometria de fluxo, independentemente de os animais terem sido submetidos à hemorragia, a dose única de parecoxibe gerou maior quantidade de necrose, menor quantidade de apoptose e maior quantidade de células viáveis.
