3.2.3 Anadolu’da Hititler Dönemi Ağırlık Ölçüleri

 A caracterização morfológica detalhada da embriogênese somática em B.

brizantha a partir de sementes maduras foi demonstrada.

 Foi obtido um sistema eficiente para micropropagação a longo prazo de

Brachiaria.

 Um sistema alternativo para embriogênese somática de sementes foi obtido usando segmentos basais de plantas cultivadas in vitro.

 Foi estabelecida uma metodologia para obtenção, multiplicação e regeneração de suspensões celulares de B. brizantha.

 Foi obtido um sistema de expressão transiente de genes para testar vetores e construções para gramíneas via biobalística usando sementes maduras.

 Calos embriogênicos bombardeados originaram plântulas resistentes a higromicina, tendo sido confirmada a presença do gene hptII por PCR, sem a presença do gene gus.

 Foi estabelecido um sistema de agroinfiltração eficiente, simples, de baixo custo e rápido para expressão transiente de genes para as cultivares Primavera de

Oryza sativa e Marandu de B. brizantha.

 Suspensões celulares de B. brizantha são suscetíveis à transformação genética por Agrobacterium tumefaciens, e foram transformadas pelas cepas EHA105 e LBA4404, pela primeira vez. Unidades embriogênicas resistentes a higromicina e expressando estavelmente o gene gus foram obtidas.

 A higromicina parece estar impedindo o desenvolvimento de proembriões resistentes ao antibiótico.

 A cultivar Primavera de Oryza sativa é recalcitrante à cultura de tecidos. O sistema de embriogênese somática via unidades embriogênicas foi caracterizado, e demonstrou ser ineficiente na regeneração, sendo necessário o aperfeiçoamento da metodologia.

 A linhagem EHA105 é capaz de transformar células de unidades embriogênicas da cultivar Primavera de Oryza sativa e a expressão da proteína marcadora GFP foi detectada.
