陳彥州教授研究分享

在這一期學術短波專欄中,編輯部請到陳彥州教授,簡介他近期的研究成果。陳教授的文章是以血紅素 化 酵素(Heme oxygenase-1; HO-1)為研究對象,經由研究推測HO-1具有潛力成為藥物開發及臨床應用之標的 基因。(編按)

血紅素 化酵素(Heme oxygenase-1; HO-1)是體內重要的抗 化壓力蛋白質之一,分子量為32kDa。人類

HO-1基因(hmox-1)位於染色體22q12位置,主要可受到金屬離子、過 化物、發炎激素等刺激物透過活

化antioxidant response element (ARE)所誘導產生,由於HO-1可受到熱休克反應活化,又稱為heat

shock protein 32 (Hsp32)。HO-1具有相當多的生理活性包括抗自由基、抗發炎和神經保護作用,臨床研究

報告也指出誘導HO-1表現能抑制血管增生、降低血管粥狀硬化、心血管疾病、糖尿病及減少器官移植排 斥等。HO-1活化後會將heme分解代謝成為膽綠素、膽紅素、鐵離子以及一 化 。膽綠素和膽紅素為體內 的抗 化劑,可保護細胞免於抗自由基造成的傷害與細胞凋亡。一 化 則能透過細胞內不同訊息傳遞路徑 如cGMP與p38MAPK等調控血管舒張、抑制發炎反應和保護細胞作用等。在我們一系列的研究中發現,HO-1 能清除體內自由基、保護神經細胞免於過 化物所造成細胞凋亡和抑制巨噬細胞發炎反應,最近我們更證 實HO-1能抑制乳癌細胞侵犯和轉移的能力,其作用機轉與抑制MMP-9基因活化有關。一些抗癌物質 如quercetin、curcumin、baicalein與sulforaphane等能透過活化HO-1基因而降低細胞癌化與抑制腫瘤生長。 因此推測HO-1具有潛力成為藥物開發及臨床應用之標的基因,進一步證實HO-1作用分子機轉與開發有效HO-1誘導劑為未來研究主要方向之一。 References:
